ACTH fragment raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
This page was last updated on 2026-06-02 and is reviewed periodically as new material appears.
Regulatory status differs sharply between jurisdictions. In Russia the peptide is registered as a prescription nasal preparation, while agencies such as the United States Food and Drug Administration have not approved it for any indication. Products sold elsewhere are typically labeled for laboratory research only, and such labels shift responsibility for safe handling to the purchaser. Because the same name covers pharmaceutical-grade nasal drops and bulk research powder, identity and purity documentation becomes the main practical concern when comparing sources.
Semax is a synthetic seven-amino-acid peptide whose sequence extends the ACTH(4-10) fragment with a C-terminal proline-glycine-proline tripeptide. The commonly cited sequence is Met-Glu-His-Phe-Pro-Gly-Pro, giving a molecular formula near C37H51N9O10S and a molecular weight close to 813.9 g/mol. It belongs to the broader class of synthetic ACTH fragments studied for central nervous system effects rather than for adrenal steroid stimulation. In practice the material appears as a lyophilized white powder for laboratory work or as a dilute saline solution in clinical settings.
Development is attributed to researchers at the Institute of Molecular Genetics in Moscow during the early 1980s, building on earlier Soviet work with ACTH fragments. Russian regulatory approval followed for intranasal use, and the compound has remained commercially available there for decades. Most published human data originate from Russian and, later, some Eastern European clinical reports, which are not always accessible in English translation. Outside that region the material is generally handled as a research chemical rather than a licensed medicine.
Terminology around the compound varies by source. It appears in catalogues and papers as Semax, as the heptapeptide ACTH(4-7)-Pro-Gly-Pro, and under various alphanumeric laboratory codes used by individual suppliers. These names refer to the same sequence but may imply different salt forms, purity grades, or counter-ions. Peptide databases usually list the free base mass, while product descriptions sometimes report acetate or trifluoroacetate salts with a different formula weight. Because naming conventions for research peptides are not standardised across vendors, checking the declared sequence and measured mass is more reliable than relying on a trade name alone.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its design combines the ACTH(4-7) core fragment with a C-terminal Pro-Gly-Pro extension, a modification intended to improve stability and prolong activity. The molecule is hydrophilic, carries no lipid chains or glycosylation, and has a theoretical mass just over 810 daltons in its free form. All seven residues are proteinogenic amino acids, so no non-natural building blocks appear in the backbone. A free N-terminal methionine and C-terminal proline define the unmodified parent peptide.

=== 1951–1953: DNA structure === In 1951 and 1952, together with William Cochran and Vladimir Vand, Crick assisted in the development of a mathematical theory of X-ray diffraction by a helical molecule. This theoretical result matched well with X-ray data for proteins that contain sequences of amino acids in the alpha helix conformation. Helical diffraction theory turned out to also be useful for understanding the structure of DNA. Late in 1951, Crick started working with James Watson at Cavendish Laboratory at the University of Cambridge, England. Using "Photo 51" (the X-ray diffraction results of Rosalind Franklin and her graduate student Raymond Gosling of King's College London, given to them by Gosling and Franklin's colleague Wilkins), Watson and Crick together developed a model for a helical structure of DNA, which they published in 1953. For this and subsequent work they were jointly awarded the Nobel Prize in Physiology or Medicine in 1962 with Wilkins. When Watson came to Cambridge, Crick was a 35-year-old graduate student (due to his work during WWII) and Watson was only 23, but had already obtained a PhD. They shared an interest in the fundamental problem of learning how genetic information might be stored in molecular form. Watson and Crick talked endlessly about DNA and the idea that it might be possible to guess a good molecular model of its structure. A key piece of experimentally-derived information came from X-ray diffraction images that had been obtained by Franklin, and Gosling.
== Notable users == King Chulalongkorn of Thailand (1853–1910) used the drug for a period after 1893 to relieve what may have been a mix of depression and unspecified illnesses. He was reported by his doctor to have been taking one bottle per day during July 1894 although this was reduced after this time. Montgomery Clift (1920–1966), American actor. André Gide (1869–1951) was given chloral hydrate as a boy for sleep problems by a physician named Lizart. Gide states in his autobiography If It Die... that "all my later weaknesses of will or memory I attribute to him." William James (1842–1910), psychologist and philosopher, used the drug for insomnia and sedation due to chronic neurosis. The Jonestown mass murder-suicides in 1978 involved the communal drinking of Flavor Aid poisoned with diazepam, chloral hydrate, cyanide, and promethazine. Mary Todd Lincoln (1818–1882), wife of American president Abraham Lincoln, became addicted in the years after her husband's death and was committed to an asylum. Marilyn Monroe (1926–1962) died from an overdose of chloral hydrate and pentobarbital (Nembutal). Friedrich Nietzsche (1844–1900) regularly used chloral hydrate in the years leading up to his nervous breakdown, according to Lou Salomé and other associates. Whether the drug contributed to his insanity is a point of controversy. Dante Gabriel Rossetti (1828–1882) became addicted to chloral, with whisky chasers, after the death of his wife Elizabeth Siddal from a laudanum overdose in 1862. He had a mental breakdown in 1872.
Work-related roadway crashes are the leading cause of death from traumatic injuries in the U.S. workplace. They accounted for nearly 12,000 deaths between 1992 and 2000. Deaths and injuries from these roadway crashes result in increased costs to employers and lost productivity in addition to their toll in human suffering. Truck drivers tend to endure higher fatality rates than workers in other occupations, but concerns about motor vehicle safety in the workplace are not limited to those surrounding the operation of large trucks. Workers outside the motor carrier industry routinely operate company-owned vehicles for deliveries, sales and repair calls, client visits, etc. In these instances, the employer providing the vehicle generally plays a major role in setting safety, maintenance, and training policy. As in non-occupational driving, young drivers are especially at risk. In the workplace, 45% of all fatal injuries to workers under age 18 between 1992 and 2000 in the United States resulted from transportation incidents.
Uniform and genetically stable cultivars can be inadequate for dealing with environmental fluctuations and novel stress factors. Plant breeders have focused on identifying crops which will ensure crops perform under these conditions; a way to achieve this is finding strains of the crop that is resistance to drought conditions with low nitrogen. It is evident from this that plant breeding is vital for future agriculture to survive as it enables farmers to produce stress resistant crops hence improving food security. In countries that experience harsh winters such as Iceland, Germany and further east in Europe, plant breeders are involved in breeding for tolerance to frost, continuous snow-cover, frost-drought (desiccation from wind and solar radiation under frost) and high moisture levels in soil in winter. Breeding is not a quick process, which is especially important when breeding to ameliorate a disease. The average time from human recognition of a new fungal disease threat to the release of a resistant crop for that pathogen is at least twelve years.

4 On 21 December 1995, a major reshuffle took place amongst existing Ministers, mainly to discharge particular Ministers of portfolios which had proven troublesome for the government. 5 On 26 April 1996, George Cash resigned from the ministry due to ill health. Graham Kierath and Kevin Minson adopted his portfolios, whilst Norman Moore was elected leader of the Government in the Legislative Council four days later.
Brent ( ) is a borough in west and north-west London, England; it forms part of Outer London. It is known for landmarks such as Wembley Stadium, the Swaminarayan Temple and the Kiln Theatre. It also contains the Welsh Harp reservoir and the Park Royal commercial estate. The local authority is Brent London Borough Council. Brent's population was estimated to be 353,000 as of 2024. Major districts are Kenton, Kilburn, Willesden, Wembley and Harlesden, with sub-districts Stonebridge, Kingsbury, Kensal Green, Neasden, and Queen's Park. Brent has a mixture of residential, industrial and commercial land. However this claim has been challenged by another report that the human musk-recognizing receptor, OR5AN1 that robustly responds to cyclopentadecanone and muscone, fails to distinguish isotopomers of these compounds in vitro. Furthermore, the mouse (methylthio)methanethiol-recognizing receptor, MOR244-3, as well as other selected human and mouse olfactory receptors, responded similarly to normal, deuterated, and carbon-13 isotopomers of their respective ligands, paralleling results found with the musk receptor OR5AN1. Hence it was concluded that the proposed vibration theory does not apply to the human musk receptor OR5AN1, mouse thiol receptor MOR244-3, or other olfactory receptors examined. In addition, the proposed electron transfer mechanism of the vibrational frequencies of odorants could be easily suppressed by quantum effects of nonodorant molecular vibrational modes. Hence multiple lines of evidence argue against the vibration theory of smell. This later study was criticized since it used "cells in a dish rather than within whole organisms" and that "expressing an olfactory receptor in human embryonic kidney cells doesn't adequately reconstitute the complex nature of olfaction...". In response, the authors of the second study state "Embryonic kidney cells are not identical to the cells in the nose .. but if you are looking at receptors, it's the best system in the world."
== Etymology == Moscow's name is thought to be derived from that of the Moskva River. Multiple theories have been proposed for the origin of this river's name. The linguistically best-grounded and most widely accepted theory is that the name comes from the Proto-Balto-Slavic root *mŭzg-/muzg-, from the Proto-Indo-European *meu- "wet"; thus, the name Moskva might signify a river at a wetland or marsh. The name's cognates include the following:

=== Differential diagnosis === IBM is often initially misdiagnosed as polymyositis. A course of prednisone is typically completed with no improvement and eventually, sIBM is confirmed. sIBM weakness comes on over months or years and progresses steadily, whereas polymyositis has an onset of weeks or months. Muscular dystrophy (e.g., limb girdle muscular dystrophy) must be considered as well. sIBM can be mistaken for physical deconditioning. Hereditary myopathies can mimic sIBM, both in signs and symptoms and in the appearance of muscle biopsies. A small percentage of those initially diagnosed with sIBM are later found to have pathogenic mutations in the genes VCP and SQSTM1, which are known to cause hIBM. IBM has a distinctive pattern of muscle involvement that distinguishes it among inflammatory myopathies. Characteristic of IBM is weakness of finger flexion, knee extension, and ankle dorsiflexion. Other inflammatory myopathies cause a proximal muscle weakness pattern, such as weakness of hip flexion, abduction, and extension, as well as shoulder abduction. IBM and other inflammatory myopathies both cause bicep/tricep weakness.
To ensure early detection, all individuals with MCTD must have screening echocardiography and high-resolution computed tomography upon diagnosis. Mild cases require regular testing to monitor for progression. Traditional therapies such as calcium channel blockers, ACE inhibitors, immunosuppression, and heart failure medications can be used. Pericarditis is typically treated with NSAIDs and/or corticosteroids based on severity. For moderate to severe myocarditis, high-dose steroid therapy should be combined with standard congestive heart failure treatment. Treatment for gastrointestinal problems in MCTD is identical to that for systemic sclerosis. First-line treatment for chronic reflux symptoms includes proton-pump inhibitors, H2-receptor antagonists, lifestyle changes, and oesophageal PH monitoring. Kidney involvement can lead to nephrotic syndrome, which may be treated with high-dose corticosteroid therapy. Corticosteroids are used to treat nervous system involvement in low-dose oral, high-dose oral, or high-dose intravenous regimens, depending on the severity of the potential harm.
== Philanthropy == Through the Pritzker Family Foundation, Pritzker has funded research and programs focused on children in poverty. Under the leadership of economist James Heckman, he supported the creation of the Pritzker Consortium on Early Childhood Development at the University of Chicago. With the Bill & Melinda Gates Foundation, the Buffett Early Childhood Fund, the Irving Harris Foundation, and the George Kaiser Family Foundation, the Pritzker Family Foundation is a founding supporter of the First Five Years Fund, an organization focusing nationwide attention and resources on comprehensive, quality early care and learning programs for children from birth to age five. In 2013, Pritzker worked with Goldman Sachs to fund the first-ever social impact bond for early childhood education. As chairman of the Illinois Holocaust Museum and Education Center, which opened in 2009, Pritzker led the capital campaign and planning to build an international institution in the Midwest dedicated to teaching the lessons of the Holocaust and other genocides. He is the principal funder of Cambodia Tribunal Monitor, the most significant online source for news and commentaries on the international criminal tribunal created to bring to justice the perpetrators of Pol Pot-era acts of genocide. He chaired the Illinois Human Rights Commission, and was succeeded by former White House counsel and federal judge Abner J. Mikva. In 2013, Pritzker received the Survivors' Legacy Award for his leadership in the creation of the Illinois Holocaust Museum and Education Center.

It is a short synthetic peptide built from seven amino acids: methionine, glutamic acid, histidine, phenylalanine and three prolines. The sequence derives from the 4-10 fragment of adrenocorticotropic hormone with an added proline-glycine-proline tail. No plant or animal extract is involved; the material is produced by solid-phase peptide synthesis.
It was developed in the Soviet Union during the early 1980s by groups at the Institute of Molecular Genetics in Moscow. Russian approval for intranasal use followed, and it has been marketed there since. Western laboratories encountered it mainly through translated literature and, later, through online research-chemical trade.
It holds a Russian registration as a prescription nasal product but has no approval from the European Medicines Agency or the United States Food and Drug Administration. Buyers outside Russia usually receive material sold strictly for laboratory research. That distinction matters because research-grade and pharmaceutical-grade products carry different documentation and testing expectations.
It is a synthetic seven-amino-acid peptide built from the ACTH(4-7) sequence plus a Pro-Gly-Pro tail. It belongs to the family of short ACTH fragments studied for nervous-system effects. Its backbone contains only standard amino acids.
